Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000122.2(ERCC3):c.1962G>C (p.Glu654Asp), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This sequence change replaces glutamic acid, which is acidic and polar, with aspartic acid, which is acidic and polar, at codon 654 of the ERCC3 protein (p.Glu654Asp). This variant is present in population databases (rs375848554, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with ERCC3-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
