Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004836.7(EIF2AK3):c.989A>C (p.Glu330Ala), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid, which is acidic and polar, with alanine, which is neutral and non-polar, at codon 330 of the EIF2AK3 protein (p.Glu330Ala). This variant is present in population databases (rs370390181, gnomAD 0.03%). This variant has not been reported in the literature in individuals affected with EIF2AK3-related conditions. ClinVar contains an entry for this variant (Variation ID: 2419088). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
