This variant lies in the FBN1 gene (transcript NM_000138.5) at coding-DNA position 213, where G is replaced by A; at the protein level this means converts the codon for tryptophan at residue 71 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This sequence change creates a premature translational stop signal (p.Trp71*) in the FBN1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in FBN1 are known to be pathogenic (PMID: 17657824, 19293843). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with Marfan syndrome (PMID: 31098894). In at least one individual the variant was observed to be de novo.
