NM_001177316.2(SLC34A3):c.1483G>A (p.Gly495Arg) was classified as Uncertain significance by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 495 of the SLC34A3 protein (p.Gly495Arg). This variant is present in population databases (rs367896891, gnomAD 0.01%). This missense change has been observed in individual(s) with clinical features of hypophosphatemic rickets with hypercalciuria and/or SLC34A3-related conditions (PMID: 31352694; Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
