Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006269.2(RP1):c.71dup (p.Arg25fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant has not been observed in the literature in individuals with autosomal recessive RP1-related conditions. This variant has been reported in individual(s) with autosomal dominant RP1-related conditions (PMID: 30913292); however, the role of the variant in this condition is currently unclear. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Arg25Serfs*37) in the RP1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in RP1 are known to be pathogenic (PMID: 11960024, 19933189).
