NM_053025.4(MYLK):c.1271A>T (p.Glu424Val) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E424V variant (also known as c.1271A>T), located in coding exon 7 of the MYLK gene, results from an A to T substitution at nucleotide position 1271. The glutamic acid at codon 424 is replaced by valine, an amino acid with dissimilar properties, and is located in the Ig-like C2-type 3 domain. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
