NM_025114.3(CEP290):c.6819-?_*171+?dup was classified as Uncertain significance for Joubert syndrome; Meckel-Gruber syndrome; Nephronophthisis by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross duplication of the genomic region encompassing exons 50-54 of the CEP290 gene. The 5' boundary is likely confined to intron 49. The 3' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. A duplication of exons 50-54 in CEP290 has not been reported in the population databases or in the published literature. In summary, the 3' boundary, genomic location and functional effect of this duplication has not been established. Therefore, it has been classified as a Variant of Uncertain Significance.
