Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000784.4(CYP27A1):c.1048C>T (p.His350Tyr), citing Ambry Variant Classification Scheme 2023: The p.H350Y variant (also known as c.1048C>T), located in coding exon 6 of the CYP27A1 gene, results from a C to T substitution at nucleotide position 1048. The histidine at codon 350 is replaced by tyrosine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
