Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001129.5(AEBP1):c.626C>T (p.Pro209Leu), citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (no rsID available, gnomAD 0.002%). This sequence change replaces proline, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 209 of the AEBP1 protein (p.Pro209Leu). This variant has not been reported in the literature in individuals affected with AEBP1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The leucine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function.
