Pathogenic for Ehlers-Danlos syndrome, kyphoscoliotic type 1 — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000302.4(PLOD1):c.1261dup (p.Ala421fs), citing LabCorp Variant Classification Summary - May 2015: Variant summary: PLOD1 c.1261dupG (p.Ala421GlyfsX45) results in a premature termination codon, predicted to cause absence of the protein due to nonsense mediated decay, which is a commonly known mechanism for disease. The variant allele was found at a frequency of 4e-06 in 251374 control chromosomes. To our knowledge, no occurrence of c.1261dupG in individuals affected with PLOD1-related conditions and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 2413497). Based on the evidence outlined above, the variant was classified as pathogenic.
