Uncertain significance for Developmental and epileptic encephalopathy, 1; Autosomal recessive spinocerebellar ataxia 12 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_016373.4(WWOX):c.101A>G (p.Tyr34Cys), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the WWOX gene (transcript NM_016373.4) at coding-DNA position 101, where A is replaced by G; at the protein level this means replaces tyrosine at residue 34 with cysteine — a missense variant. Submitter rationale: In summary, this variant is a novel missense change with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance. This sequence change replaces tyrosine with cysteine at codon 34 of the WWOX protein (p.Tyr34Cys). There is a large physicochemical difference between tyrosine and cysteine. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a WWOX-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies.
