Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007194.4(CHEK2):c.827T>G (p.Ile276Ser), citing Ambry Variant Classification Scheme 2023: The p.I276S variant (also known as c.827T>G), located in coding exon 6 of the CHEK2 gene, results from a T to G substitution at nucleotide position 827. The isoleucine at codon 276 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
