NM_007194.4(CHEK2):c.25G>A (p.Ala9Thr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A9T variant (also known as c.25G>A), located in coding exon 1 of the CHEK2 gene, results from a G to A substitution at nucleotide position 25. The alanine at codon 9 is replaced by threonine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
