Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_005862.3(STAG1):c.1597A>G (p.Ile533Val), citing Ambry Variant Classification Scheme 2023: The c.1597A>G (p.I533V) alteration is located in exon 16 (coding exon 15) of the STAG1 gene. This alteration results from a A to G substitution at nucleotide position 1597, causing the isoleucine (I) at amino acid position 533 to be replaced by a valine (V). Based on data from gnomAD, the G allele has an overall frequency of 0.001% (2/236970) total alleles studied. The highest observed frequency was 0.007% (1/15214) of African alleles. This amino acid position is highly conserved in available vertebrate species. This alteration is predicted to be tolerated by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
