Uncertain significance for Dilated cardiomyopathy 1O — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020297.4(ABCC9):c.3095A>G (p.Gln1032Arg), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a ABCC9-related disease. This sequence change replaces glutamine with arginine at codon 1032 of the ABCC9 protein (p.Gln1032Arg). The glutamine residue is moderately conserved and there is a small physicochemical difference between glutamine and arginine. In summary, this is a novel missense change with uncertain impact on splicing. It has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies. However, algorithms developed to predict the effect of nucleotide changes on mRNA splicing suggest that this variant may alter mRNA splicing, but this prediction has not been confirmed by published transcriptional studies.
