Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_004260.4(RECQL4):c.3573C>T (p.Ser1191=), citing Sema4 Curation Guidelines: The RECQL4 c.3573C>T (p.S1191=) variant has not been reported in the literature to our knowledge. It was observed in 33/10272 chromosomes of the Ashkenazi Jewish subpopulation, with no homozygotes, in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has been reported in ClinVar (Variation ID 239767). The position is moderately conserved and in silico tools suggest that the variant does not impact splicing, though these predictions have not been confirmed by functional studies. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
