Pathogenic for Pheochromocytoma/paraganglioma syndrome 3; Gastrointestinal stromal tumor — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003001.3(SDHC):c.406-?_*2318+?del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon 6 of the SDHC gene. The 5' boundary is likely confined to intron 5. The 3' end of this event is unknown as it extends through the termination codon beyond the assayed region for this gene and may encompass additional genes. While this deletion is not anticipated to result in nonsense mediated decay, it is expected to create a truncated SDHC protein. A deletion encompassing exon 6 has been reported to segregate with disease in one family affected with head and neck paragangliomas (PMID: 15342702). In the same study, it was also found in an independent individual affected with the same condition but not in any of the 103 control individuals tested (PMID: 15342702). For these reasons, this variant has been classified as Pathogenic.
