Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001232.4(CASQ2):c.361C>T (p.Arg121Cys), citing Ambry Variant Classification Scheme 2023: The p.R121C variant (also known as c.361C>T), located in coding exon 3 of the CASQ2 gene, results from a C to T substitution at nucleotide position 361. The arginine at codon 121 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
