NM_001134363.3(RBM20):c.247C>A (p.Leu83Ile) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the RBM20 gene (transcript NM_001134363.3) at coding-DNA position 247, where C is replaced by A; at the protein level this means replaces leucine at residue 83 with isoleucine — a missense variant. Submitter rationale: The p.L83I variant (also known as c.247C>A), located in coding exon 2 of the RBM20 gene, results from a C to A substitution at nucleotide position 247. The leucine at codon 83 is replaced by isoleucine, an amino acid with highly similar properties. This variant was reported in one individual from a dilated cardiomyopathy (DCM) cohort; however, clinical details were limited (Refaat MM et al. Heart Rhythm, 2012 Mar;9:390-6). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
