NM_006940.6(SOX5):c.131G>A (p.Ser44Asn) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.131G>A (p.S44N) alteration is located in exon 2 (coding exon 2) of the SOX5 gene. This alteration results from a G to A substitution at nucleotide position 131, causing the serine (S) at amino acid position 44 to be replaced by an asparagine (N). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is well conserved in available vertebrate species. This alteration is predicted to be tolerated by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
