However, the evidence is insufficient at this time to prove that conclusively. It has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies. This missense change is located within a functionally conserved Kinase domain of the STK11 protein (PMID: 16707622 and 85% of previously reported STK11 missense and truncating mutations have been found within this domain (PMID: 16707622). These observations suggest that a novel missense substitution within this domain may affect protein function, but experiments have not been done to test this possibility. This variant has been reported in an individual affected with Peutz-Jeghers syndrome (PMID:11389158). Different missense substitutions at this codon (p.Gly242Glu) and (p.Gly242Val) are reported in individuals with STK11-related disorders (PMID:11389158,12865922,16707622). This indicates that the glycine residue is important for STK11 protein function. This variant is not present in population databases (ExAC no frequency). This sequence change replaces glycine with tryptophan at codon 242 of the STK11 protein (p.Gly242Trp). The glycine residue is highly conserved and there is a large physicochemical difference between glycine and tryptophan.
