Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001009944.3(PKD1):c.8281C>T (p.Arg2761Cys), citing LabCorp Variant Classification Summary - May 2015: Variant summary: PKD1 c.8281C>T (p.Arg2761Cys) results in a non-conservative amino acid change located in the REJ domain (IPR014010) of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 0.00021 in 247570 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in PKD1 causing Polycystic Kidney Disease 1 (0.00021 vs 0.0005), allowing no conclusion about variant significance. c.8281C>T has been reported in the literature in an individual affected with Polycystic Kidney Disease who also had a second variant of uncertain significance in PKD1 (Kurashige_2015). These data do not allow any conclusion about variant significance. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
