This variant lies in the EYS gene (transcript NM_001142800.2) at coding-DNA position 6137, where G is replaced by A; at the protein level this means converts the codon for tryptophan at residue 2046 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The p.Trp2046Ter variant in EYS was identified in an individual with Retinitis pigmentosa, via a collaborative study between the Broad Institute's Center for Mendelian Genomics and the Pierce lab (https://oculargenomics.meei.harvard.edu/labs/pierce-lab/lab-members/).
