Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000059.4(BRCA2):c.6588_6589del (p.Lys2196fs), citing Ambry Variant Classification Scheme 2023: The c.6588_6589delAA pathogenic mutation, located in coding exon 10 of the BRCA2 gene, results from a deletion of two nucleotides at nucleotide positions 6588 to 6589, causing a translational frameshift with a predicted alternate stop codon (p.K2196Nfs*2). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This variant is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this variant is interpreted as a disease-causing mutation.
