Uncertain significance — the classification assigned by Ambry Genetics to NM_012282.4(KCNE5):c.223G>A (p.Gly75Arg), citing Ambry Variant Classification Scheme 2023: The p.G75R variant (also known as c.223G>A), located in coding exon 1 of the KCNE5 gene, results from a G to A substitution at nucleotide position 223. The glycine at codon 75 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
