This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 109, where T is replaced by G; at the protein level this means replaces serine at residue 37 with alanine — a missense variant. Submitter rationale: Variant summary: BRCA2 c.109T>G (p.Ser37Ala) results in a conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be tolerated. The variant was absent in 251120 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.109T>G in individuals affected with BRCA2-related conditions has been reported. At least one publication reports experimental evidence evaluating an impact on protein function. These results showed no damaging effect of this variant (Bose_2025).
