This variant lies in the MYH7 gene (transcript NM_000257.4) at coding-DNA position 635, where G is replaced by A; at the protein level this means replaces glycine at residue 212 with aspartic acid — a missense variant. Submitter rationale: The G212D variant has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge. This variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. Missense variants in nearby residues (K207Q, S210R, P211L, G214D, L216V, D218Y) have been reported in the Human Gene Mutation Database in association with HCM (Stenson et al., 2014), supporting the functional importance of this region of the protein. The G212D variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties.
