Uncertain significance — the classification assigned by Quest Diagnostics Nichols Institute San Juan Capistrano to NM_000179.3(MSH6):c.3350G>A (p.Cys1117Tyr), citing Quest Diagnostics criteria. This variant lies in the MSH6 gene (transcript NM_000179.3) at coding-DNA position 3350, where G is replaced by A; at the protein level this means replaces cysteine at residue 1117 with tyrosine — a missense variant. Submitter rationale: The MSH6 c.3350G>A (p.Cys1117Tyr) variant has been reported in the published literature in an individual with colorectal cancer (PMID: 29245953 (2017)), and in a reportedly unaffected individual (PMID: 33471991 (2021), see also LOVD (http://databases.lovd.nl/shared/)). This variant has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded conflicting predictions that this variant is benign or damaging. Based on the available information, we are unable to determine the clinical significance of this variant.
