Submitter rationale: This variant is denoted CHEK2 c.371G>T at the cDNA level, p.Cys124Phe (C124F) at the protein level, and results in the change of a Cysteine to a Phenylalanine (TGC>TTC) in exon 3. This variant has not, to our knowledge, been published in the literature as either a mutation or a benign polymorphism. CHEK2 Cys124Phe was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. Since Cysteine and Phenylalanine differ in polarity, charge, size or other properties, this is considered a non-conservative amino acid substitution. CHEK2 Cys124Phe occurs at a position that is not conserved and is located in the FHA domain (Desrichard 2011, Roeb 2012).
