NM_033198.4(PIGS):c.294G>A (p.Met98Ile) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.294G>A (p.M98I) alteration is located in exon 4 (coding exon 4) of the PIGS gene. This alteration results from a G to A substitution at nucleotide position 294, causing the methionine (M) at amino acid position 98 to be replaced by an isoleucine (I). Based on data from the Genome Aggregation Database (gnomAD) database, the PIGS c.294G>A alteration was observed in <0.001% (1/251476) of total alleles studied, with a frequency of 0.003% (1/34592) in the Latino subpopulation. This amino acid position is well conserved in available vertebrate species. The p.M98I alteration is predicted to be tolerated by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
