NM_000179.3(MSH6):c.3979A>C (p.Asn1327His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N1327H variant (also known as c.3979A>C), located in coding exon 9 of the MSH6 gene, results from an A to C substitution at nucleotide position 3979. The asparagine at codon 1327 is replaced by histidine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
