This sequence change replaces threonine, which is neutral and polar, with proline, which is neutral and non-polar, at codon 26 of the PTEN protein (p.Thr26Pro). This missense change has been observed in individual(s) with clinical features of PTEN hamartoma tumor syndrome (PMID: 16773562, 21194675, 22595938). In at least one individual the variant was observed to be de novo. For these reasons, this variant has been classified as Pathogenic. This variant disrupts the p.Thr26 amino acid residue in PTEN. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 25549896; Invitae). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. Experimental studies have shown that this missense change affects PTEN function (PMID: 29706633).
