NM_005732.4(RAD50):c.2129A>G (p.Asp710Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D710G variant (also known as c.2129A>G), located in coding exon 13 of the RAD50 gene, results from an A to G substitution at nucleotide position 2129. The aspartic acid at codon 710 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
