Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007294.4(BRCA1):c.2156A>C (p.Lys719Thr), citing Ambry Variant Classification Scheme 2023: The p.K719T variant (also known as c.2156A>C), located in coding exon 9 of the BRCA1 gene, results from an A to C substitution at nucleotide position 2156. The lysine at codon 719 is replaced by threonine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear.
