NM_000051.4(ATM):c.2264G>T (p.Cys755Phe) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 2264, where G is replaced by T; at the protein level this means replaces cysteine at residue 755 with phenylalanine — a missense variant. Submitter rationale: The p.C755F variant (also known as c.2264G>T), located in coding exon 14 of the ATM gene, results from a G to T substitution at nucleotide position 2264. The cysteine at codon 755 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6499 samples (12998 alleles) with coverage at this position. To date, this alteration has been detected with an allele frequency of approximately 0.002% (greater than 66000 alleles tested) in our clinical cohort. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of p.C755F remains unclear.
