NM_003000.3(SDHB):c.80G>A (p.Arg27Gln) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R27Q variant (also known as c.80G>A), located in coding exon 2 of the SDHB gene, results from a G to A substitution at nucleotide position 80. The arginine at codon 27 is replaced by glutamine, an amino acid with highly similar properties. This alteration has been reported in a patient diagnosed with a pheochromocytoma at age 44 (D&eacute;nes J et al. J. Clin. Endocrinol. Metab., 2015 Mar;100:E531-41). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
