Uncertain Significance for Hereditary pheochromocytoma and paraganglioma — the classification assigned by All of Us Research Program, National Institutes of Health to NM_003000.3(SDHB):c.80G>A (p.Arg27Gln), citing ACMG Guidelines, 2015. This variant lies in the SDHB gene (transcript NM_003000.3) at coding-DNA position 80, where G is replaced by A; at the protein level this means replaces arginine at residue 27 with glutamine — a missense variant. Submitter rationale: This missense variant replaces arginine with glutamine at codon 27 of the SDHB protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in individuals affected with pheochromocytoma and or paraganglioma in the literature (PMID: 25494863, 34906457). This variant has been identified in 5/250678 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
