This variant lies in the ATM gene (transcript NM_000051.4) at the canonical splice donor site of the intron immediately after coding-DNA position 4236, where G is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: ATM c.4236+1G>T is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Several computational tools predict a significant impact on normal splicing: Three predict the variant abolishes a 5 splicing donor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 248804 control chromosomes (gnomAD). c.4236+1G>T has been reported in the literature in individuals affected with ovarian cancer, lung adenocarcinoma and renal cell carcinoma (e.g. Carter_2018, Esai Selvan_2020, Truong_2021). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Four ClinVar submitters (evaluation after 2014) cite the variant as likely pathogenic.
