This variant lies in the RAD50 gene (transcript NM_005732.4) at coding-DNA position 3373, where T is replaced by A; at the protein level this means replaces tyrosine at residue 1125 with asparagine — a missense variant. Submitter rationale: The p.Y1125N variant (also known as c.3373T>A), located in coding exon 21 of the RAD50 gene, results from a T to A substitution at nucleotide position 3373. The tyrosine at codon 1125 is replaced by asparagine, an amino acid with dissimilar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6466 samples (12932 alleles) with coverage at this position. To date, this alteration has been detected with an allele frequency of approximately 0.002% (greater than 65000 alleles tested) in our clinical cohort. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of p.Y1125N remains unclear.
