Submitter rationale: This missense variant replaces tyrosine with cysteine at codon 72 of the CHEK2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. In a large case-control study, this variant has shown inconclusive association with breast cancer (2/60464 cases, 1/53460 controls, OR=1.768, 95%CI 0.16 to 19.503; PMID 33471991). This variant has been identified in 5/251474 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
