Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005732.4(RAD50):c.2891A>G (p.Tyr964Cys), citing Ambry Variant Classification Scheme 2023: The p.Y964C variant (also known as c.2891A>G), located in coding exon 18 of the RAD50 gene, results from an A to G substitution at nucleotide position 2891. The tyrosine at codon 964 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
