Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_004100.5(EYA4):c.191C>G (p.Thr64Ser), citing Ambry Variant Classification Scheme 2023: The c.191C>G (p.T64S) alteration is located in exon 4 (coding exon 3) of the EYA4 gene. This alteration results from a C to G substitution at nucleotide position 191, causing the threonine (T) at amino acid position 64 to be replaced by a serine (S). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is highly conserved in available vertebrate species. This alteration is predicted to be tolerated by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
