NM_000051.4(ATM):c.6301C>G (p.Gln2101Glu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q2101E variant (also known as c.6301C>G), located in coding exon 42 of the ATM gene, results from a C to G substitution at nucleotide position 6301. The glutamine at codon 2101 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
