NM_007294.4(BRCA1):c.533T>A (p.Val178Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Color Diagnostics, LLC DBA Color Health, citing ACMG Guidelines, 2015. This variant lies in the BRCA1 gene (transcript NM_007294.4) at coding-DNA position 533, where T is replaced by A; at the protein level this means replaces valine at residue 178 with aspartic acid — a missense variant. Submitter rationale: This missense variant replaces valine with aspartic acid at codon 178 of the BRCA1 protein. Computational prediction suggests that this variant may have deleterious impact on protein structure and function (internally defined REVEL score threshold >= 0.7, PMID: 27666373). Splice site prediction tools suggest that this variant may not impact RNA splicing. To our knowledge, functional studies have not been performed for this variant. This variant has been reported in an individual affected with breast cancer, as well as in a healthy control individual (PMID: 28993434). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
