Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_004329.3(BMPR1A):c.560G>A (p.Arg187His), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the BMPR1A gene (transcript NM_004329.3) at coding-DNA position 560, where G is replaced by A; at the protein level this means replaces arginine at residue 187 with histidine — a missense variant. Submitter rationale: Variant summary: BMPR1A c.560G>A (p.Arg187His) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 251316 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. The variant has been reported in an individual affected with breast cancer (Tung_2016), however, to our knowledge, no occurrence of c.560G>A in individuals affected with Juvenile Polyposis Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. Four clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation, and classified the variant as VUS (n=3) or likely benign (n=1). Based on the evidence outlined above, the variant was classified as uncertain significance.
