NM_003001.5(SDHC):c.431A>C (p.Lys144Thr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K144T variant (also known as c.431A>C), located in coding exon 6 of the SDHC gene, results from an A to C substitution at nucleotide position 431. The lysine at codon 144 is replaced by threonine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
