Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_017617.5(NOTCH1):c.1951G>T (p.Asp651Tyr), citing Ambry Variant Classification Scheme 2023: The p.D651Y variant (also known as c.1951G>T), located in coding exon 12 of the NOTCH1 gene, results from a G to T substitution at nucleotide position 1951. The aspartic acid at codon 651 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
