This variant lies in the CDH1 gene (transcript NM_004360.5) at coding-DNA position 158, where G is replaced by C; at the protein level this means replaces glycine at residue 53 with alanine — a missense variant. Submitter rationale: The p.G53A variant (also known as c.158G>C), located in coding exon 2 of the CDH1 gene, results from a G to C substitution at nucleotide position 158. The glycine at codon 53 is replaced by alanine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 5321 samples (10642 alleles) with coverage at this position. To date, this alteration has been detected with an allele frequency of approximately 0.001% (greater than 105000 alleles tested) in our clinical cohort. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be possibly damaging and tolerated by PolyPhen and SIFT in silico analyses, respectively. Since supporting evidence is limited at this time, the clinical significance of p.G53A remains unclear.
