NM_007194.4(CHEK2):c.707T>A (p.Leu236Gln) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L236Q variant (also known as c.707T>A), located in coding exon 5 of the CHEK2 gene, results from a T to A substitution at nucleotide position 707. The leucine at codon 236 is replaced by glutamine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
