Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007194.4(CHEK2):c.974A>T (p.Lys325Met), citing Ambry Variant Classification Scheme 2023: The p.K325M variant (also known as c.974A>T), located in coding exon 8 of the CHEK2 gene, results from an A to T substitution at nucleotide position 974. The lysine at codon 325 is replaced by methionine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
