This variant lies in the FH gene (transcript NM_000143.4) at coding-DNA position 92, where C is replaced by T; at the protein level this means replaces alanine at residue 31 with valine — a missense variant. Submitter rationale: The p.A31V variant (also known as c.92C>T), located in coding exon 1 of the FH gene, results from a C to T substitution at nucleotide position 92. The alanine at codon 31 is replaced by valine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 4951 samples (9902 alleles) with coverage at this position. To date, this alteration has been detected with an allele frequency of approximately 0.01% (greater than 20000 alleles tested) in our clinical cohort. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
